硫代辅酶I的新人工合成设计

硫代辅酶I(SNAD)是一种重要的试剂原料,具有高克分子消光系数和高稳定性的优点,具有替代NAD的趋势,可以广泛应用在酶活力测定和代谢物测定,以提高测定体系的灵敏度和精密度。它的人工合成至今为止尚未提出。
从市售的腺苷(adenosine)开始,得到I (5'- 二苯胺基磷酰基-腺苷),它与从市售的硫代烟酰胺(thionicotinamide)开始得到的II(3'位由(CH3O)C(CH3)2保护和硫酰胺基由(BOC)2O保护的的5'-二苯硫基磷酰基-核糖-硫代烟酰胺)结合得到硫代辅酶I。

Abstract:
Thio NAD I (SNAD) is an important reagent raw materials, with the advantages of high molar extinction coefficient and high stability, with a trend to alternate NAD, can be widely used in the enzyme activity assay and the determination of metabolites, in order to improve the sensitivity and precision of the determinated system. Its synthetic routes so far have not yet to be submitted.
I (5'-diphenylamine phosphoryl - adenosine), which can be obtained from the commercially available AMP (adenosine) , was combined with II (5'-diphenylthio phosphoryl-ribose-thionicotinamide, wherein, 3' was protected by (CH3O)C(CH3)2 and sulfur amide was protected by (BOC) 2O), which can be obtained from the commercially available thio-nicotinamide, to get thio NAD I.
